Autograft- one location to another

Acute rejection (days to weeks after transplant)
Chronic rejection (months to years)- vessel wall damage, smooth muscle proliferation and vessel occlusion.
Hyperacute rejection (minutes to hours)- pre formed antibodies (cause)
GHD- bone marrow (or to a lesser extent liver)- recipient who has received them- proliferating cell- result is rash, jaundice, diarrhea and gastrointestinal haemorrhage
ABO blood grouping and cross matching (if we have no time)- cross matching involves mixing the cells of the donor with serum from the recipient- mixing erythrocytes with known sera.
persons with type O- anti A and anti B isohemagglutinins- universal donor
A blood have A antigens- make anti-B antibodies
B blood have B antigens- make Anti A antibodies
AB blood make no isohemagglutinins have A and B antigens- universal recipients
HLA matching of donor and recipients- A,B and DR (DR is best if have to choose)- match to class II- this may seem counterintuitive
The mixed lymphocyte reaction (class II testing)
MHC II stimulation - production of IL2- cells from donor - cells from recipient are added in culture- source of radiolabelled nucleotide precursor- newly formed DNA radioactivity of culture to rise- best result would be no increase in radioactivity
Class I testing (microcytotoxicity or flow cytometry)
microcytotoxicity- treat the cells of donor and recipient - very large stockpile of specific antisera - focus can be achieved by common haplotypes in particular ethnic groups- complement add- cell membrane become leaky- add color dye- leaky membranes are identified
